Diff-Quik Stain Kit Introduction
迪夫快速染色液(Diff-Quik)是在瑞氏 - 吉姆萨染色基础上改良而成的快速罗曼诺夫斯基染色试剂。相较于传统瑞氏染色、瑞氏 - 吉姆萨染色,该试剂可在短时间内完成细胞分化染色,大幅缩短实验耗时。
迪夫染色液包含三种工作液:甲醇固定液、伊红酸性染液、亚甲蓝 - 天青 A 碱性染液。细胞内嗜酸性物质与伊红结合,呈现红粉色;细胞核等嗜碱性结构与噻嗪类染料结合,显蓝紫色。本品染色背景干净,细胞形态清晰,分色效果稳定,适用于实验室大批量快速检测。
Diff-Quik is a rapid modified Romanowsky staining reagent improved based on Wright-Giemsa stain. It can complete cell differential staining in a short time, greatly saving experimental time compared with traditional Wright staining and Wright-Giemsa staining.
The kit contains three reagents: methanol fixative, eosin-based acidic dye solution and methylene blue-azure A alkaline dye solution. Acidophilic substances in cells combine with eosin to show red-pink; basophilic structures such as cell nuclei bind to thiazine dyes to present blue-purple. The staining background is clean, cell morphology is clear, and the color separation effect is stable, which is suitable for rapid batch detection in laboratories.

血液检测:血涂片、骨髓涂片,白细胞分类计数
Blood examination: blood smears, bone marrow smears, leukocyte differential count
脱落细胞病理:穿刺细胞、痰液、胸腹水、阴道分泌物涂片
Cytopathology: fine needle aspiration cells, sputum, pleural and ascitic fluid, vaginal discharge smears
男科检测:精子形态染色(WHO 精液分析标准染色方法)
Andrology test: sperm morphology staining (WHO standard for semen evaluation)
微生物与动物实验:腹腔灌洗液、组织印片、细胞爬片快速染色
Microbiology & animal experiments: peritoneal lavage fluid, tissue touch preps, cell slide staining
四、操作步骤
4.1 涂片自然风干
Air-dry the cell smear completely at room temperature.
4.2 固定:浸入 Diff-Quik 固定液 20 s
Immerse slide in Diff-Quik Fixative for 20 seconds for cell fixation.
4.3 染 A 液:浸泡 / 上下涮洗 5–10 s,甩掉多余染液
Stain with Solution Ⅰ for 5–10 seconds, drain excess liquid.
4.14 染 B 液:浸泡 / 上下涮洗 10–20 s
Stain with Solution Ⅱ for 10–20 seconds.
4.5 水洗:蒸馏水轻柔冲洗玻片
Gently rinse slide with distilled water to wash off unbound dye.
4.6 吸水纸吸干,镜检
Blot slide dry with filter paper, observe under optical microscope.
五、染色结果
※ 细胞核、嗜碱性颗粒:蓝紫色
Cell nucleus, basophil granules: blue-purple
※ 红细胞、细胞质、基质:粉红色 / 橘红色
Red blood cells, cytoplasm, matrix: pink/salmon red
※ 单核、淋巴细胞胞质:淡紫红色
Cytoplasm of monocytes and lymphocytes: light purple
Diff-Quik Stain Kit
1. Brief Introduction
Diff-Quik stain is a rapid modified Romanowsky-type staining technique derived from Wright-Giemsa stain, officially recommended by WHO for routine cytological and hematological slide staining.
Compared with traditional Wright staining, it drastically shortens the whole staining procedure to less than 90 seconds, delivering comparable differential color discrimination with clean background and minimal precipitate, ideal for high-throughput batch staining.
The kit consists of three components: methanol-based fixative, eosin-containing acidic staining solution I, and thiazine-based alkaline staining solution II (methylene blue + Azure A).
Solution I (eosin Y, anionic dye): Stains acidophilic structures (cytoplasm, erythrocytes, cell matrix) pink-red.
Solution II (methylene blue & Azure A, cationic dyes): Stains basophilic components (cell nucleus, nucleolus, RNA-rich granules) blue-purple; metachromatic purple staining appears on lymphocyte cytoplasm and monocyte nucleiSiemens He....
2. Main Applications
Hematology: Blood smears, bone marrow smears, leukocyte differential counting
Cytopathology: Exfoliative cell smears, fine needle aspiration (FNA) specimens, vaginal discharge smears
Reproductive lab: Sperm morphology staining (WHO standard method for sperm assessment)
Histology: Frozen & paraffin section rapid screening
3. Standard Staining Protocol
Air-dry the smear naturally; immerse in Diff-Quik Fixative for 20 seconds to stabilize cellular morphology.
Dip slides up and down in Diff-Quik I for 5–10 seconds, drain excess liquid.
Dip slides up and down in Diff-Quik II for 10–20 seconds, drain excess liquid.
Rinse gently with distilled water to remove residual dye.
Blot dry with filter paper; observe under light microscope immediately. Permanent mounting with neutral balsam is optional for long-term preservation.

1.实验前需戴好防护眼镜,穿戴防护服和口罩,佩戴手套,避免与皮肤接触。
2.实验过程中如遇到有毒或者刺激性物质及有害物质产生,必要时实验操作需要手套箱内完成以免对实验人员造成伤害。
3.取样品的移液枪头需及时更换,必要时为避免交叉污染尽可能选择滤芯吸头。
4.称量药品时选用称量纸,并无风处取药和称量以免扬撒,试剂的容器使用前务必确保干净,并消毒。
5.取药品时尽量采用多个药勺分别使用,使用后清洗干净。6.
实验后产生的废弃物需分类存储,并交于专业生物废气物处理公司处理,以免造成环境污染。Experimental considerations:
